Q1. Summary of the internship (Description of the research conducted, aim, introduction of lab, and things you learned)
I completed my internship at the International Research Center for Medical Sciences (IRCMS), Kumamoto University, from January 14, 2026, to February 28, 2026. Under the expert supervision of Professor Yuta Takahashi, I engaged in research centered on epigenetic inheritance. Epigenetic inheritance refers to the transmission of information from one cell generation to the next or from parent to offspring that does not involve changes to the underlying DNA nucleotide sequence. Instead, this inheritance is mediated by molecular "tags," such as DNA methylation, which act as a regulatory layer that dictates whether genes are turned "on" or "off." By studying these processes, we can understand how cells maintain their identity and how environmental factors can lead to stable, long-term changes in gene activity without altering the genetic code itself. The primary objective of my internship was to generate locus-specific, endogenous protein-degradable knock-ins for the DNA methyltransferases DNMT1, DNMT3a, and DNMT3b using the CRISPR-Cas9 system. By integrating a BSD-P2A-dTAG selection cassette at these specific genomic loci, the study aims to establish a robust cellular model that allows for the rapid and reversible depletion of these enzymes. This approach provides a "switch-off" mechanism, enabling the precise investigation of their individual and collective roles in DNA methylation and epigenetic regulation by allowing us to study the immediate cellular consequences of their loss in real-time.

Beyond technical skills, this internship experience has significantly shaped my career trajectory by working on this project, I have gained a specialized understanding of how to manipulate epigenetic regulators to study cellular mechanisms.
